بررسی اثر عصاره گیاه شیرین بیان بر روند تنظیم ژن aflR و روند تولید آفلاتوکسین در قارچ آسپرژیلوس پارازیتیکوس به روش Real Time PCR
A Survey of the Effect of Licorice Plant Extract on aflR Gene Expression and Aflatoxin Production in Aspergillus Parasiticus via Real-time PCR
هدف: آفلاتوکسین در صنایع غذایی، دامپروری و حوزۀ پزشکی اهمیت دارد و آسیب‏های اقتصادی فراوانی را سبب می‏شود. مطالعات زیادی روی عصاره‏ها و ترکیبات گیاهی برای کاهش رشد میکروارگانیسم‏های مولد آفلاتوکسین، عدم تولید سم توسط این ارگانیسم‏ها و سرکوب نمودن ژن اصلی مولد این توکسین یعنی <em>aflR</em> در جریان است. از گیاهانی دارای اهمیت در طب سنتی ما که آثار ضد میکروبی از خود نشان داده، شیرین بیان است. هنوز گزارشی از آثار یا مکانیسم‏های اثر آن بر قارچ‏های آسپرژیلوس مولد آفلاتوکسین وجود ندارد. مطالعۀ کنونی به بررسی اثر عصاره شیرین بیان بر مهار رشد آسپرژیلوس پارازیتیکوس و بیان ژن <em>alfR</em> توسط این ارگانیسم می‏پردازد. مواد و روش‏ها: پس از کشت آسپرژیلوس پارازیتیکوس در محیط محرک تولید آفلاتوکسین، حداقل غلظت بازدارندگی از رشد قارچ تحت اثر عصارۀ شیرین بیان اندازه‏گیری شد. غلظت آفلاتوکسین در محیط‏های کنترل و تیمار شده به روشHPLC سنجیده شد. همچنین این قارچ از محیط محرک توکسین جدا و mRNA آن استخراج شد. سپس با استفاده از آغازگرهای عمومی cDNA آن سنتز و تغییر در بیان ژن <em>aflR</em> به‏طور کمّی و با استفاده از PCR در زمان واقعی بررسی شد. در نهایت تجزیه و تحلیل آماری توسط SPSS نسخه 14 صورت گرفت. نتایج: با افزایش غلظت عصاره شیرین بیان میزان تولید میسلیوم قارچ کاهش ‏یافت. بیشترین میزان مهار کنندگی از رشد قارچ در غلظت 500 میلی‏گرم بر میلی‏لیتر مشاهده شد. تجزیه و تحلیل نتایج HPLC نشان داد که مؤثرترین غلظت عصاره شیرین بیان، غلظت 10 میلی‏گرم بر میلی‏لیتر بود که باعث مهار تولید توکسین به میزان 99/99 درصد شد. سنجش کمّی بیان ژن <em>aflR</em> تحت تأثیر عصاره شیرین بیان نشان داد که بیان این ژن تا میزان 40 درصد مهار شده است. نتیجه‏گیری: به‏طور کلی عصاره شیرین بیان می تواند به‏طور چشمگیری تأثیرات مهارکنندگی بر بیان ژن <em>aflR</em> و تولید آفلاتوکسین در قارچ آسپرژیلوس پارازیتیکوس داشته باشد.
Objective: Aflatoxin is important in the food industry, in animal husbandry and the medical area; there are enormous negative economic impacts due to this toxin. Numerous studies have researched extracts and plant compounds with the intent to reduce the growth of aflatoxin-producing organisms, inhibit toxin production and suppress the major toxin encoded genes (i.e., <em>aflR</em>) in these organisms. Licorice is an important plant in traditional medicine that possesses numerous antimicrobial activities. There is no report regarding the effects of licorice or its mechanism of action on the aflatoxin-producing <em>Aspergillus</em> species. The present study focuses on the inhibitory effects of licorice extract on <em>aflR</em> gene expression and the<em> </em>growth and survival of<em> Aspergillus parasiticus (A. parasiticus)</em>. Methods: After the culture of <em>A. parasiticus</em> in toxin-inducer medium, we measured the minimal inhibitory concentration (MIC) for licorice extract. The aflatoxin concentration in the control and treated media was determined by HPLC. After harvesting the fungi from the toxin-inducing medium, its mRNA was extracted and cDNA synthesized by universal primers. The quantitative change in the <em>aflR</em> expression was analyzed via real-time PCR. Statistical analysis was performed by SPSS (v16). Results: The production of fungal mycelium decreased with increasing concentrations of licorice extract. The highest inhibitory concentration observed was 500 mg/ml of the extract. HPLC analyses revealed that the 10 mg/ml concentration of licorice extract inhibited toxin production by 99.9%. At this concentration, <em>aflR</em> gene expression was suppressed up to 40% as documented by quantitative RT-PCR analysis. Conclusion: Overall we concluded that the Licorice extract could inhibit the <em>aflR</em> gene expression and consequently the aflatoxin production efficiently in the <em>A. parasiticus.</em>
