القای اتوفاژی با ژن Beclin 1 و تأثیر آن روی نکروز رده سلولی MCDK
Induction of Autophagy by the Beclin 1 Gene and Its Effect on MDCK Cell Line Necrosis
هدف: اتوفاژی (خودخواری) یک فرآیند بسیار تنظیم شده برای تخریب پروتئینهای آسیبدیده و اجزا درون سلولی از کارافتاده است. اتوفاژی نقشهای چندگانهای در حفظ هموستازی سلول دارد. بکلین 1 (Beclin 1) یک مولکول تنظیمی بسیار مهم در شروع و شکلگیری اتوفاگوزوم است. مهار اتوفاژی بهوسیله تخریب آللی بکلین1 حساسیت به استرس متابولیک را ایجاد میکند. مهار کردن اتوفاژی تحت شرایط محدودیت مواد مغذی در تومورهای مقاوم به مرگ سلولی برنامهریزی شده، منجر به نکروز التهاب و رشد تومور افزایش یافته همراه است.<br /> هدف از این مطالعه، القای اتوفاژی با ژن بکلین 1 و تأثیر آن در روند نکروز رده سلولی MCDK بود.<br /> مواد و روشها: در این مطالعه، القای اتوفاژی با ژن بکلین 1 در سلولهای MDCK بررسی شد. سپس درصد مرگ سلولی نکروز در این سلولها با روش فلوسایتومتری با استفاده از کیت رنگآمیزی آنکسین V سنجیده شد. بهمنظور القا کردن اتوفاژی، سازه HYPERLINK "../../../../../abstract/med/21426771/" t "_blank"Beclin1-(-)pcDNA 3.1 با استفاده از لیپوفکتامین 3000 درون رده سلولی MDCK ترنسفکت شد.<br /> نتایج: در مطالعه حاضر مشاهده شد که بیان بالای ژن بکلین1 در سلولهای MDCK به القای اتوفاژی منجر میشود، همانگونه که با رنگآمیزی شاخص اتوفاگوزومی داخل سلولی دیده شد. درصد ساختارهای LC3 مثبت در سلولهای ترنسفکت شده و نشده بهترتیب 92/9 و 15/0 درصد بود. در گروههای ترنسفکت شده و کنترل، درصد مرگ سلولی نکروز، بهترتیب 66/1 و 06/0 درصد بود.<br /> نتیجهگیری: تداخل بین مسیرهای اتوفاژی و نکروز ممکن است بر سرنوشت طول عمر سلول اثر داشته باشد. استراتژیهای درگیر در تعدیل کردن اتوفاژی و مرگ سلولی ممکن است بر روند درمان بیماریها مؤثر باشد. بنابراین دستکاری مسیرهای مرگ سلولی ممکن است حوزههای جدیدی را در استفادههای درمانی و مداخلهای ایجاد کند.
Objective: Autophagy (self-digestion) is a highly regulated process for the degradation of damaged proteins and intracellular components. Autophagy has multifunctional roles in the protection of cellular homeostasis. Beclin1 is a key regulator molecule in autophagosome formation. Inhibition of autophagy by destruction of the Beclin 1 allele creates sensitivity to metabolic stress. Inhibition of the autophagy under conditions of nutrient deprivation in tumors resistant to apoptosis can lead to necrosis, inflammation and increased tumor growth. This study aims to assess the effect of autophagy induction on the necrosis pathway of MDCK cells. <br />Methods: We evaluated induction of autophagy by the Beclin1 gene in MDCK cells and assessed the percentage of necrosis cell death by flow cytometry using an Annexin V Staining kit. In order to induce autophagy, the recombinant pcDNA3.1-Beclin 1 was transfected into the MDCK cell line using lipofectamine TM 3000. <br />Results: Overexpression of the Beclin1 gene in MDCK cells led to induction of autophagy as seen by intracellular autophagosomal indicator LC3-II staining. There were 9.92% positive LC3 structures in transfected cells and 0.15% in untransfected cells. In the transfected and control groups, the rate of necrosis cell death was 1.66% and 0.06%, respectively. <br />Conclusion: Crosstalk between autophagy and necrosis pathways might affect the fate of the cell life span. Strategies that involve in modulation of autophagy and cell death might lead to therapeutic interventions in diseases. Therefore manipulation of cell death pathways could create new areas in therapeutic uses and interventions.
